Submitter rationale: The p.G266E variant (also known as c.797G>A), located in coding exon 8 of the FMR1 gene, results from a G to A substitution at nucleotide position 797. The glycine at codon 266 is replaced by glutamic acid, an amino acid with similar properties. This alteration was detected as maternally inherited in an individual with a diagnosis of Fragile X syndrome who did not carry the typical repeat expansion mutation. This alteration was not detected in this individual's three unaffected brothers. In addition, authors used in vitro assays to show that this alteration impaired several FMRP protein functions (Myrick LK et al. Eur. J. Hum. Genet., 2014 Oct;22:1185-9).
