Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001182.5(ALDH7A1):c.452T>C (p.Val151Ala), citing Ambry Variant Classification Scheme 2023. This variant lies in the ALDH7A1 gene (transcript NM_001182.5) at coding-DNA position 452, where T is replaced by C; at the protein level this means replaces valine at residue 151 with alanine — a missense variant. Submitter rationale: The p.V151A variant (also known as c.452T>C), located in coding exon 5 of the ALDH7A1 gene, results from a T to C substitution at nucleotide position 452. The valine at codon 151 is replaced by alanine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6500 samples (13000 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
