Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_007325.5(GRIA3):c.1850T>C (p.Met617Thr), citing Ambry Variant Classification Scheme 2023. This variant lies in the GRIA3 gene (transcript NM_007325.5) at coding-DNA position 1850, where T is replaced by C; at the protein level this means replaces methionine at residue 617 with threonine — a missense variant. Submitter rationale: The p.M617T variant (also known as c.1850T>C), located in coding exon 11 of the GRIA3 gene, results from a T to C substitution at nucleotide position 1850. The methionine at codon 617 is replaced by threonine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6502 samples with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
