NM_001127222.2(CACNA1A):c.1696A>C (p.Ile566Leu) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I567L variant (also known as c.1699A>C), located in coding exon 13 of the CACNA1A gene, results from an A to C substitution at nucleotide position 1699. The isoleucine at codon 567 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and leucine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
