Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001365999.1(SZT2):c.6038A>G (p.Tyr2013Cys), citing Ambry Variant Classification Scheme 2023: The p.Y1956C variant (also known as c.5867A>G), located in coding exon 42 of the SZT2 gene, results from an A to G substitution at nucleotide position 5867. The tyrosine at codon 1956 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
