Uncertain significance — the classification assigned by Ambry Genetics to NM_153026.3(PRICKLE1):c.699G>T (p.Lys233Asn), citing Ambry Variant Classification Scheme 2023: The p.K233N variant (also known as c.699G>T), located in coding exon 5 of the PRICKLE1 gene, results from a G to T substitution at nucleotide position 699. The lysine at codon 233 is replaced by asparagine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
