Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000719.7(CACNA1C):c.6068G>A (p.Gly2023Glu), citing Ambry Variant Classification Scheme 2023: The p.G2023E variant (also known as c.6068G>A), located in coding exon 46 of the CACNA1C gene, results from a G to A substitution at nucleotide position 6068. The glycine at codon 2023 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
