Uncertain significance for Emery-Dreifuss muscular dystrophy 4, autosomal dominant; Autosomal recessive ataxia, Beauce type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_182961.4(SYNE1):c.13832A>C (p.Gln4611Pro), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SYNE1 gene (transcript NM_182961.4) at coding-DNA position 13832, where A is replaced by C; at the protein level this means replaces glutamine at residue 4611 with proline — a missense variant. Submitter rationale: This sequence change replaces glutamine with proline at codon 4540 of the SYNE1 protein (p.Gln4540Pro). The glutamine residue is highly conserved and there is a moderate physicochemical difference between glutamine and proline. This variant is present in population databases (rs762392590, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with SYNE1-related conditions. ClinVar contains an entry for this variant (Variation ID: 586709). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
