Pathogenic for Retinitis pigmentosa — the classification assigned by Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard to NM_024649.5(BBS1):c.1177C>T (p.Arg393Ter), citing ACMG Guidelines, 2015. This variant lies in the BBS1 gene (transcript NM_024649.5) at coding-DNA position 1177, where C is replaced by T; at the protein level this means converts the codon for arginine at residue 393 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The p.Arg393Ter variant in BBS1 was identified in an individual with Retinitis pigmentosa, via a collaborative study between the Broad Institute's Center for Mendelian Genomics and the Pierce lab (https://oculargenomics.meei.harvard.edu/labs/pierce-lab/lab-members/). Through a review of available evidence we were able to apply the following criteria: PVS1, PM2, PM3-P. Based on this evidence we have classified this variant as Pathogenic. If you have any questions about the classification please reach out to the Pierce Lab.
