Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000059.4(BRCA2):c.9337A>G (p.Ile3113Val), citing ACMG Guidelines, 2015: This missense variant replaces isoleucine with valine at codon 3113 of the BRCA2 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function. Functional studies have reported that this variant does not impact BRCA2 in a haploid cell proliferation assay and in sensitivity assays to cisplatin and PARP inhibitor (PMID: 39779848, 39779857). This variant has been reported in an individual affected with breast cancer, and it has been detected in a breast cancer case-control meta-analysis in 1/60466 cases and 1/53461 unaffected individuals (PMID: 33471991, 35264596Leiden Open Variation Database DB-ID BRCA2_008754). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
