Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.431T>C (p.Met144Thr), citing Ambry Variant Classification Scheme 2023: The p.M144T variant (also known as c.431T>C), located in coding exon 4 of the RECQL gene, results from a T to C substitution at nucleotide position 431. The methionine at codon 144 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
