Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_058216.3(RAD51C):c.912T>G (p.Ser304Arg), citing Ambry Variant Classification Scheme 2023: The p.S304R variant (also known as c.912T>G), located in coding exon 7 of the RAD51C gene, results from a T to G substitution at nucleotide position 912. The serine at codon 304 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
