The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 1 of the PMS2 gene. This is expected to result in an absent or disrupted protein product. Loss-of-function variants in PMS2 are known to be pathogenic. A deletion of exon 1 has been reported in the literature in an individual with colorectal cancer (PMID: 18809606, 18602922), and was observed in trans with a second pathogenic PMS2 variant (c.24-12_107del96insAAAT) in two siblings with phenotypes consistent with constitutional mismatch repair-deficiency (PMID: 24068316). For these reasons, this variant has been classified as Pathogenic.
