Pathogenic for Familial cancer of breast — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.10:g.(?_23623999)_(23626407_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 7-8 of the PALB2 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in PALB2 are known to be pathogenic (PMID: 17200668, 24136930, 25099575). This variant has been observed in an individual affected with breast cancer (PMID:¬†24136930).
