Pathogenic for Pontocerebellar hypoplasia type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000014.9:g.(?_96846075)_(96847364_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 4-5 of the VRK1 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. This variant has not been reported in the literature in individuals with VRK1-related disease. Loss-of-function variants in VRK1 are known to be pathogenic (PMID: 19646678). For these reasons, this variant has been classified as Pathogenic.
