NC_000002.12:g.(?_32114618)_(32147278_?)del was classified as Pathogenic for Hereditary spastic paraplegia 4 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 5-16 of the SPAST gene. This is expected to create a premature translational stop signal, while this is not anticipated to result in nonsense mediated decay, it is expected¬†to result in a disrupted protein product. A similar deletion of exons 5-16 has been reported in an individual affected with spastic paraplegia (PMID: 25065914). This deletion encompasses a portion of the microtubule binding domain and the entire ATPase Associated with various cellular Activities (AAA) domain. A significant number of previously reported missense mutations have been found within these domains (PMID: 10699187, 18701882, 11809724). These observations suggest that a deletion of these domains will also affect protein function. For these reasons, this variant has been classified as Pathogenic.
