NC_000006.12:g.(?_24527977)_(24528186_?)del was classified as Pathogenic for Succinate-semialdehyde dehydrogenase deficiency by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the ALDH5A1 protein in which other variant(s) (p.Tyr519*) have been determined to be pathogenic (PMID: 23430864). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with ALDH5A1-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 8 of the ALDH5A1 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to disrupt the C-terminus of the protein.
