Pathogenic for Arrhythmogenic right ventricular cardiomyopathy, type 9 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000012.12:g.(?_32824025)_(32824182_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exon 8 of the PKP2 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. A similar deletion of exon 8 has been reported in an individual affected with arrhythmogenic right ventricular cardiomyopathy who also has a variant in DSC2 (PMID:Â¬â€ 21606396).Â¬â€ ClinVar contains an entry for this variant (Variation ID:Â¬â€ 164945). Loss-of-function variants in PKP2 are known to be pathogenic (PMID: 15489853, 23911551). For these reasons, this variant has been classified as Pathogenic.
