Pathogenic for Hereditary Breast Carcinoma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000022.11:g.(?_28724967)_(28734731_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 2-4 of the CHEK2 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 4 of the CHEK2 gene. This is expected to result in an absent or disrupted protein product. A deletion of exons 2-4 has been observed in an individual undergoing hereditary cancer testing (PMID: 24763289). Loss-of-function variants in CHEK2 are known to be pathogenic (PMID: 21876083, 24713400). For these reasons, this variant has been classified as Pathogenic.
