Pathogenic for Dilated cardiomyopathy with woolly hair and keratoderma; Arrhythmogenic right ventricular cardiomyopathy, type 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000006.12:g.(?_7570417)_(7585898_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 13-24 of the DSP gene. The 5' boundary is likely confined to intron 12. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated protein product or disrupt mRNA translation. This variant has not been reported in the literature in individuals with DSP-related disease. Truncations (p.Arg2166* and p.Thr2634Lysfs*4) that lie within the deleted region of this variant have been determined to be pathogenic (PMID: 26899768,Â¬â€ 11063735). This suggests that deletion of this region of the DSP protein is causative of disease. For these reasons, this variant has been classified as Pathogenic.
