Pathogenic for Neurofibromatosis, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.11:g.(?_31155977)_(31163382_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an in-frame deletion of the genomic region encompassing exons 2-11 of the NF1 gene. It preserves the integrity of the reading frame. This variant has not been reported in the literature in individuals with NF1-related disease. Several variants located in this region (p.Leu380Pro, p.Ser82Phe, p.Arg160Thr, and p.Val253del) have been reported in individuals affected with neurofibromatosis type 1 and have been determined to be likely pathogenic or pathogenic (PMID: 27999334, 21520333, 11857752, 17311297, 21838856, Invitae). This suggests that exons 2-11 is critical for NF1 protein function and deletion of this region may also be pathogenic. For these reasons, this variant has been classified as Pathogenic.
