Pathogenic for Neurofibromatosis, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.11:g.(?_31169871)_(31343155_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). While this particular deletion has not been reported in the literature in individuals with NF1-related disease, gross deletions of the NF1 gene have been observed in many individuals with neurofibromatosis type 1 (PMID: 8116612, 8931693, 9643287). This variant is an out-of-frame deletion of the genomic region encompassing exons 5-47 of the NF1 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product.
