NC_000015.9:g.(?_26792920)_(27018162_?)dup was classified as Uncertain significance for Epilepsy, childhood absence 5; Epilepsy, childhood absence 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant results in a copy number gain of the genomic region encompassing the full coding sequence of the GABRB3 gene. The boundaries of this event are unknown as they extend beyond the assayed region for this gene and therefore may encompass additional genes. As the precise location of this event is unknown, it may be in tandem or it may be located elsewhere in the genome. While a whole-gene copy number gain of just the GABRB3 gene alone has not been reported in the literature, much larger copy number variants encompassing more than 5 Mb of DNA (15q11-q13) and multiple adjacent genes have been reported in individuals with a wide spectrum of neuropsychiatric disorders, including autism spectrum disorders, developmental delay, learning difficulties, schizophrenia, and seizures (PMID: 26068938, 23495136, 2828157). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
