Pathogenic for Hereditary nonpolyposis colon cancer — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000002.12:g.(?_47373463)_(47377077_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 2-7 of the EPCAM gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. A similar deletion has not been reported in the literature in individuals with EPCAM-related disease. Loss-of-function variants in EPCAM are known to be pathogenic (PMID: 21315192, 20034091). For these reasons, this variant has been classified as Pathogenic.
