Uncertain significance for Citrin deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014251.3(SLC25A13):c.614C>T (p.Ala205Val), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces alanine with valine at codon 205 of the SLC25A13 protein (p.Ala205Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. This variant has not been reported in the literature in individuals with SLC25A13-related disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
