The 5' end of this event is located 44 nucleotides upstream of the initiator codon and the 3' boundary is confined to intron 1 of the ERF gene (c.-44_22+11del). This is expected to result in an absent or disrupted protein product. While this variant is not present in population databases, the frequency information is unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has been observed in an individual affected with craniosynostosis (Invitae). However, it has also been observed in one or more individuals who were not affected with craniosynostosis (Invitae).
