NM_000492.4(CFTR):c.634A>G (p.Met212Val) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M212V variant (also known as c.634A>G), located in coding exon 6 of the CFTR gene, results from an A to G substitution at nucleotide position 634. The methionine at codon 212 is replaced by valine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
