This variant lies in the RANBP2 gene (transcript NM_006267.5) at coding-DNA position 8411, where G is replaced by T; at the protein level this means replaces serine at residue 2804 with isoleucine — a missense variant. Submitter rationale: This sequence change replaces serine with isoleucine at codon 2804 of the RANBP2 protein (p.Ser2804Ile). The serine residue is highly conserved and there is a large physicochemical difference between serine and isoleucine. This variant is present in population databases (rs368049213, ExAC 0.03%). This variant has not been reported in the literature in individuals with RANBP2-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
