Pathogenic for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.6700C>T (p.Gln2234Ter), citing Ambry Variant Classification Scheme 2023. This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 6700, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 2234 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The p.Q2213* variant (also known as c.6637C>T), located in coding exon 43 of the NF1 gene, results from a C to T substitution at nucleotide position 6637. This changes the amino acid from a glutamine to a stop codon within coding exon 43. This alteration has been observed in a Turkish infant with CALMs and a first degree relative with NF1 diagnosis (Hazan F et al. Neurol Sci, 2021 May;42:2045-2057). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
