This variant lies in the DAG1 gene (transcript NM_004393.6) at coding-DNA position 23, where C is replaced by T; at the protein level this means replaces serine at residue 8 with leucine — a missense variant. Submitter rationale: This sequence change replaces serine with leucine at codon 8 of the DAG1 protein (p.Ser8Leu). The serine residue is weakly conserved and there is a large physicochemical difference between serine and leucine. This variant is present in population databases (rs199501149, ExAC 0.005%). This variant has not been reported in the literature in individuals affected with DAG1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
