This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 5993, where A is replaced by C; at the protein level this means replaces glutamic acid at residue 1998 with alanine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid with alanine at codon 2025 of the PLEC protein (p.Glu2025Ala). The glutamic acid residue is highly conserved and there is a moderate physicochemical difference between glutamic acid and alanine. This variant is present in population databases (rs782286124, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with PLEC-related conditions.
