Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 0.00012 in 251490 control chromosomes, predominantly at a frequency of 0.0015 within the African or African-American subpopulation in the gnomAD database. This frequency is not significantly higher than estimated for disease-causing variants in RNASEH2A, allowing no conclusion about variant significance. To our knowledge, no occurrence of c.374T>C in individuals affected with RNASEH2A-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
