Uncertain significance for Spermatogenic failure 18; Ciliary dyskinesia, primary, 37 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015512.5(DNAH1):c.7150A>C (p.Asn2384His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine with histidine at codon 2384 of the DNAH1 protein (p.Asn2384His). The asparagine residue is weakly conserved and there is a small physicochemical difference between asparagine and histidine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with DNAH1-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
