Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003239.5(TGFB3):c.301T>C (p.Tyr101His), citing Ambry Variant Classification Scheme 2023: The p.Y101H variant (also known as c.301T>C), located in coding exon 1 of the TGFB3 gene, results from a T to C substitution at nucleotide position 301. The tyrosine at codon 101 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
