This variant lies in the SMAD4 gene (transcript NM_005359.6) at coding-DNA position 1612, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 538 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change results in a premature translational stop signal in the SMAD4 gene (p.Glu538*). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 15 amino acids of the SMAD4 protein. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
