NM_001754.5(RUNX1):c.408T>A (p.Asn136Lys) was classified as Uncertain significance for Hereditary thrombocytopenia and hematological cancer predisposition syndrome associated with RUNX1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine with lysine at codon 136 of the RUNX1 protein (p.Asn136Lys). The asparagine residue is moderately conserved and there is a moderate physicochemical difference between asparagine and lysine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in an individual affected with chronic myelomonocytic leukemia (PMID: 25840971). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have shown that this missense change impairs the normal function of RUNX1 protein (PMID: 25840971).
