NM_007194.4(CHEK2):c.1552A>G (p.Ser518Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S518G variant (also known as c.1552A>G), located in coding exon 14 of the CHEK2 gene, results from an A to G substitution at nucleotide position 1552. The serine at codon 518 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
