Submitter rationale: The APC c.6781C>T (p.P2261S) variant has not been reported in the literature to our knowledge. It was observed in 1/251062 chromosomes in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654), and has been reported in ClinVar (Variation ID 581185). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive.
