Pathogenic for Mendelian susceptibility to mycobacterial diseases due to complete IL12RB1 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005535.3(IL12RB1):c.251_254del (p.Gly84fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in IL12RB1 are known to be pathogenic (PMID: 9603733, 12591909). This variant has not been reported in the literature in individuals with IL12RB1-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Gly84Alafs*45) in the IL12RB1 gene. It is expected to result in an absent or disrupted protein product.
