This variant lies in the DICER1 gene (transcript NM_177438.3) at coding-DNA position 5261 through 79 bases into the intron immediately after coding-DNA position 5364, deleting this region. Submitter rationale: This variant is a gross deletion of the genomic region encompassing part of exon 24 of the DICER1 gene including exon 24-intron 24 boundary (c.5261_5364+79del). This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). Similar partial deletions have not been reported in the literature in individuals with DICER1-related disease. Loss-of-function variants in DICER1 are known to be pathogenic (PMID: 19556464, 21266384). For these reasons, this variant has been classified as Pathogenic.
