Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.537+5A>G, citing Ambry Variant Classification Scheme 2023: The c.537+5A>G intronic variant results from an A to G substitution 5 nucleotides after coding exon 5 in the PMS2 gene. In silico splice site analysis predicts that this alteration will not have any significant effect on splicing; however, direct evidence is insufficient at this time (Ambry internal data). This nucleotide position is poorly conserved in available vertebrate species. Based on the available evidence, the clinical significance of this variant remains unclear.
