Submitter rationale: This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 4190 of the RYR2 protein (p.Val4190Met). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal dominant catecholaminergic polymorphic ventricular tachycardia (internal data). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 580536). Other variant(s) that disrupt this residue have been observed in individuals with RYR2-related conditions (internal data), which suggests that this may be a clinically significant amino acid residue. For these reasons, this variant has been classified as Pathogenic.
