Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.00027 in 224686 control chromosomes. This frequency is not significantly higher than estimated for disease-causing variants in TBK1, allowing no conclusion about variant significance. c.1957G>C has been observed in individual(s) affected with frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS), as well as in the control subjects (vanderzee_2017). These report(s) do not provide unequivocal conclusions about association of the variant with Autoinflammation with arthritis and vasculitis. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
