Uncertain significance for Aortic aneurysm, familial thoracic 7 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_053025.4(MYLK):c.1651+1G>A, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects a donor splice site in intron 12 of the MYLK gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with MYLK-related disease. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant occurs in the long isoform of MYLK (PMID: 21055718). The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in the long isoform of MYLK cause disease.
