Uncertain significance for Combined immunodeficiency due to OX40 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003327.4(TNFRSF4):c.146-2A>G, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects an acceptor splice site in intron 1 of the TNFRSF4 gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with TNFRSF4-related disease. The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in TNFRSF4 cause disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
