Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.2130C>A (p.Asp710Glu), citing Ambry Variant Classification Scheme 2023: The p.D710E variant (also known as c.2130C>A), located in coding exon 14 of the PTCH1 gene, results from a C to A substitution at nucleotide position 2130. The aspartic acid at codon 710 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
