Uncertain significance for Charcot-Marie-Tooth disease type 2E — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006158.5(NEFL):c.58G>A (p.Glu20Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid with lysine at codon 20 of the NEFL protein (p.Glu20Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with NEFL-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleteriois"; PolyPhen-2: "Not Available"; Align-GVGD: "Class C15").
