Loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). This variant has been reported in individuals in the Leiden Open-source Variation Database (PMID: 21520333). In addition, a different variant (c.7727C>G) giving rise to the same protein effect observed here (p.Ser2576*) has been reported in an individual affected with neurofibromatosis type 1 (PMID: 18041031). This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Ser2576*) in the NF1 gene. It is expected to result in an absent or disrupted protein product.
