This variant lies in the CACNA1H gene (transcript NM_021098.3) at coding-DNA position 4316, where C is replaced by T; at the protein level this means replaces alanine at residue 1439 with valine — a missense variant. Submitter rationale: This sequence change replaces alanine with valine at codon 1439 of the CACNA1H protein (p.Ala1439Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with CACNA1H-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
