Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006231.4(POLE):c.5174-2A>C, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the POLE gene (transcript NM_006231.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 5174, where A is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 38 of the POLE gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with POLE-related conditions. ClinVar contains an entry for this variant (Variation ID: 578973). Studies have shown that disruption of this splice site results in skipping of exon 39, and produces a non-functional protein and/or introduces a premature termination codon (internal data). For these reasons, this variant has been classified as Pathogenic.
